Objective 

To assess the effect of serum obtained from women with APLS on invasion capacity and MMP2 and MMP9 secretion of JAR human choriocarcinoma cells.

Study Design 

Blood was collected from 12 APLS and 10 healthy non-pregnant women. APLS was diagnosed and confirmed in women with a history of recurrent miscarriages, late fetal death or thrombosis, and with positive anticardiolipin antibodies and/or lupus anticoagulant at least twice 12 weeks apart. Controls were healthy women with a normal obstetric history and a negative thrombophilia workup. The JAR human choriocarcinoma cell line was cultured in the presence of 10% serum. Cell invasion and MMP (2 and 9) secretion were measured using the Matrigel invasion assay, and gel zymography, respectively.

Results 

JAR invasion was reduced by 21% in the APLS group (68+5%) compared to controls (89+6%) (P<0.01). MMP-2 secretion was reduced in the APLS group by 69% (319+127 study vs 1029+149 controls optical density units, P<0.03). MMP-9 secretion was not significantly different between the groups (228+11 study vs 637+183 controls optical density units, P=0.07).

Conclusion 

Serum obtained from women with APLS has an inhibitory effect on JAR trophoblast cell invasiveness and MMP-2 secretion. This effect may be involved in the early pathologic implantation associated with the pathogenesis of APLS.
